5'-Nucleotidase of rat liver microsomes.

چکیده
Recent observations from this laboratory have revealed that there is an alteration in the kinetic properties of microsomal glucose 6-phosphatase in the livers of diabetic rats (1, Z), in addition to the’well established increase in total activity (3, 4). Since the kinetic differences between the normal and the diabetic enzyme disappeared upon disruption of the microsomes (Z), it seemed possible that they arose from alterations in the microsomes themselves. In order to obtain more information related to this possibility, an examination of a second microsomal enzyme was undertaken. The phosphatase which hydrolyzes adenosine 5’-phosphate (5, 6) was selected for this purpose. No changes were observed in either the kinetics or activity of thii enzyme in diabetic animals. Pursuant to these studies the specificity and a number of the properties of the enzyme were investigated and are reported herein.

A procedure has been developed for the cytochemical localization of 5'-nucleotidase in isolated, unfixed, rat liver microsomes. Membranes were incubated with adenosine 5'-phosphate (5'-AMP) and Pb(NO 3)2 and then isolated on sucrose density gradients : all the phosphate released was recovered with the membranes by this procedure . Adenosine 2'phosphate (2'-AMP) and adenosine 3',5'-cyclic phosph...

متن کاملSubcellular localization and properties of 5'-nucleotidase in the rat liver.
By means of procedures for isolating subcellular structural components in a state free from cross-contamination, 5’nucleotidase of the rat liver has been localized to the membranous component of the microsomes. Evidence has been obtained that suggests that a large fraction of the 5’-nucleotidase activity resides in the plasma membrane fragments that constitute a part of the population of the mi...

Preparations enriched with plasmalemmal, outer mitochondrial, or Golgi complex membranes from rat liver were subfractionated by isopycnic centrifugation, without or after treatment with digitonin, to establish the subcellular distribution of a variety of enzymes. The typical plasmalemmal enzymes 5'-nucleotidase, alkaline phosphodiesterase I, and alkaline phosphatase were markedly shifted by dig...
